My guidesmen 500 lumen flashlight takes these rechargeable batteries but the light seems to run on a non-rechargeable cr123 as well. Will this damage the flashlight in any way?
We highly reccomend using the approved rechargeable battery. It is very possible to damage the light using anything else.

What side of this battery is neg and which is positive? One side has a blue/green ring.
Hi. If there isn't a symbol on the side of the battery somewhere, typically one end is raised slightly which is the positive terminal. Otherwise the side with the ring is positive.

Why does it have the different terminals on it verses other rechargeable batteries with the same specs?
The most critical thing assuming all specs are equal is the length.
